چکیده

The present study determined the ability of the Lactobacillus rhamnosus GG strain (LGG) to enhance the anticancer effects of geniposide on HSC-3 human oral squamous carcinoma cells. LGG (1.0x103 CFU/ml) on its own had no impact on human oral keratinocytes and HSC-3 cancer cells. Geniposide (25 or 50 μg/ml) had no impact on human oral keratinocytes, but exerted growth inhibitory effects on HSC-3 cancer cells, which were increased in the presence of LGG. Flow cytometric analysis and a nuclear staining assay with DAPI revealed that HSC-3 cancer cells treated with LGG-geniposide (1.0x103 CFU/ml LGG and 50 μg/ml geniposide) had a higher apoptotic rate than cells in other treatment groups, particularly that treated with geniposide (50 μg/ml) only. Geniposide also increased the mRNA and protein expression of caspase-3, -8 and -9 as well as B-cell lymphoma 2 (Bcl-2)-associated X protein, p53, p21, inhibitor of nuclear factor-κB (NF-κB) α, Fas and Fas ligand, while decreasing Bcl-2, Bcl extra large protein, inhibitor of apoptosis-1 and -2, NF-κB, cyclooxigenase-2 and inducible nitric oxide synthase in HSC-3 cells, which was increased in the presence of LGG. These results indicated that LGG enhanced the anticancer effects of geniposide in HSC-3 cells.

The Inhibitory Effect of Lactobacillus rhamnosus GG on Vancomycin Resistant Enterococcus faecalis Colonization in Mouse

Background&Aims: Vancomycin Resistant Enterococci (VRE) are among the most common nosocomial pathogens worldwide. The intestinal tract provides a major source for transmission of these bacteria. Probiotics are living microorganisms that moderate use of them has inhibitory effect on intestinal colonization by enteric pathogens. We examined the effect of Lactobacillus rhamnosus GG (LGG) on inhibi...

Preliminary evaluation of Lactobacillus rhamnosus strain GG, a potential probiotic in dogs.

Lactobacillus rhamnosus strain GG (LGG) has been studied extensively as a probiotic in humans. However, the ability of an organism to survive passage through the intestinal tract and exert beneficial effects cannot be directly extrapolated between species. This study evaluated the ability of LGG to survive gastrointestinal transit in dogs and assessed whether oral administration of LGG is safe,...

Probiotic Lactobacillus rhamnosus GG and Aloe vera gel improve lipid profiles in hypercholesterolemic rats.

OBJECTIVE The effects of Lactobacillus rhamnosus GG (LGG) and Aloe vera (AV) gel on lipid profiles in rats with induced hypercholesterolemia were studied. METHODS Five treatment groups of rats (n = 7) were the fed experimental diets: a normal control diet, a hypercholesterolemic diet (HD), HD + LGG, HD + AV gel, and HD + LGG + AV gel. RESULTS Supplementation with LGG decreased serum total c...

Antifungal defense of probiotic Lactobacillus rhamnosus GG is mediated by blocking adhesion and nutrient depletion

Candida albicans is an inhabitant of mucosal surfaces in healthy individuals but also the most common cause of fungal nosocomial blood stream infections, associated with high morbidity and mortality. As such life-threatening infections often disseminate from superficial mucosal infections we aimed to study the use of probiotic Lactobacillus rhamnosus GG (LGG) in prevention of mucosal C. albican...

Immunomodulation of Lactobacillus rhamnosus GG (LGG)-derived soluble factors on antigen-presenting cells of healthy blood donors

Lactobacillus rhamnosus GG (LGG) cells have been shown to promote type-1 immune responsiveness; however knowledge of immunomodulation of soluble factors secreted by LGG is limited. This is the first study to investigate whether LGG soluble factors promote a comparable immune responsiveness as the bacterial cells. Both treatments - LGG conditioned medium with (CM + LGG) or without (CM) LGG cells...
